ABSTRACT

Three-dimensional (3D) organoid culture systems emerge as potential reliable tools for studying the basic developmental processes of human diseases, in particular cancer. The present study used established and modified culture conditions to report the successful generation and characterization of patient-derived organoids from fresh primary tissue samples from patients with naive prostate cancer (PCa). processing. Fresh tissue samples were collected, enzymatically digested and the resulting cell suspensions were plated in a 3D environment using Matrigel as an extracellular matrix. The 12-factor medium previously established for the organoid culture was modified to create a minimal 5-factor medium. Organoids and corresponding tissue samples were characterized using transcriptomic analysis, immunofluorescence analysis, and immunohistochemistry. In addition, organoids derived from the patient were used to assess the response to the drug. PCa organoids derived from treatment naïve patients were obtained from fresh radical prostatectomy specimens. These PCa organoids mimicked the heterogeneity of the corresponding parental tumor tissue. Histopathological analysis demonstrated similar tissue architecture and cell morphology, as well as consistent immunohistochemical marker expression. In addition, the results confirmed the potential of organoids as in vitro model to assess potential personalized therapeutic responses, as there was a differential drug response between different patient samples. In conclusion, the present study examined organoids derived from a cohort of treatment-naïve patients. The derived organoids mimicked the histological features and prostate lineage profiles of their corresponding parental tissue and could present a potential model for predicting patient-specific treatment response in a preclinical setting.

The House of Representatives approved at second reading the measure to create a separate institution for high-level offenders.

Bill 10355 or the High Level Offenders Separate Facilities Act was approved in a voice vote in Wednesday’s plenary session.

It aims to establish and maintain a separate, secure and hygienic penitentiary for the custody of high-level offenders serving time at the facilities of the Bureau of Corrections.

The bill defines high-level offenders as those convicted of heinous crimes and identified by the CEO of BuCor as high-risk or high-profile, taking into account the offense for which the inmate is convicted, the record judicial or inmate background, and affiliations.

The location of the institution will be determined by the Secretary of Justice and it should be isolated from the general population and other detainees.

The bill said it would be better if the facility was located in a military establishment or on an island separate from the mainland. He added that Luzon, Visayas and Mindanao should have separate facilities.

Senate President Vicente Sotto III earlier proposed that those considered high-level drug traffickers and other heinous criminals be separated from other convicts, noting that there should be a national penitentiary for them. – Anna Felicia Bajo / RSJ, GMA News

When Kingsley Amis met the works of DH Lawrence, he was not impressed through yet another great denouncer and missionary, the English send themselves “to tell them themselves that they are rude, rude, lost, dead, mad and addicted to unnatural vice.” With little charity, Amis suggested leaving the didactic novelist on “his prime, inspiring, unreachable and unread.”

Leaving aside questions of inspiration, the same can be said of Hillary Clinton, ousted US presidential candidate, cranky alarmist in office, still worried about establishment power incursions by rogue elements eager to seize the crown of power. One of them, in his mind, is the threat posed by cryptocurrency.

During a panel discussion at the Bloomberg New Economy Forum in Singapore, she made the following comment is worth quoting in its entirety: “Another area that I hope nation states start to pay more attention to is the rise of cryptocurrency, because what seems like a very interesting effort and somewhat exotic to literally mine new coins to trade with them has the potential to undermine currencies, to undermine the dollar’s role as a reserve currency, to destabilize nations, perhaps starting with small but going much larger. “

The remark has every threat imaginable in the Clinton universe: the potential dethronement of the almighty dollar, the cornerstone and guarantee of American power; the attack on the nation-state; false exoticism. For other establishment figures, cryptocurrencies are a regulatory nightmare, which can be used for money laundering, human trafficking and drug trafficking. The fact that all of this goes very smoothly with standard currency transactions is another one of those troublesome truths that doesn’t disrupt the dystopian terror tale.

Other politicians are also nervous about breakthroughs in crypto. Indian Prime Minister Narendra Modi, imbued with his own brand of creepy paternalism, told a forum organized by the Australian Strategic Policy Institute on November 18, that it was “important that all democratic nations work together on this issue and make sure this does not end in the wrong hands, which can ruin our young people.”

Such a mood was not helped by the Indian Supreme Court ruling in March last year, effectively overturning a 2018 central bank order banning banks from trading cryptocurrencies. The decision saw a meteoric investment boom.

Without being discouraged, a government official Modi Recount Reuters in March that a new bill was under consideration that would ban cryptocurrencies and criminalize the possession, issuance, mining, trading and transfer of those assets. The target here is private crypto assets; the preference goes to blockchain technology.

Despite the severity of this bill, Finance Minister Nirmala Sitharaman was also throw some crumbs to reassure worried investors. “I can only give you this clue that we are not closing our minds, we are looking at ways in which experiences can occur in the digital world and cryptocurrency.”

El Salvador has already carved out a niche in the jungle of nation states for crypto, give it legal tender in September. Until mid-November, the country hosted Bitcoin Week, and the Minister of the Economy, María Luisa Hayem, was excited to say at the Adopting Bitcoin conference, how his government “committed to innovate” and expressed pride in embracing digital currency. Rosily, she proclaims that “the currency gave, in a short time, access to payments and services that Salvadorans did not have before”.

The movement had been more than frowned upon by these agents of poverty, the world Bank and the International Monetary Fund, the latter warning against “macroeconomic, financial and legal issues that require very careful analysis” arising from the adoption of such currencies. But even central banks, threatened by this decentralizing digital march, are doing the trick, yet there is a problem that these deposits could exacerbate financial instability.

In Australia, the Commonwealth Bank ventured into the crypto market in partnership with digital currency trading platform Gemini and blockchain analytics firm Chainalysis, development an application allowing customers to “buy, sell and hold crypto assets”. Ten crypto assets will include, among them Bitcoin, Bitcoin Cash, Litecoin, and Ethereum.

The gesture is far from cavalier, and even a little boring. The CBA wants a piece of the crypto pie and thinks it can reassure customers that it can do so in a regulated system. Have a dedicated exchange platform will mean that digital currencies from other exchanges will not be allowed to be introduced. Close monitoring is promised.

Clinton’s fears and warnings of unscrupulous digital barbarians going on a rampage find themselves in cold isolation. The current cryptocurrency market is worth $ 2,000 billion, which is remarkable considering that crypto only came into being in 2009. The market capitalization of digital assets, according to the numbers from JP Morgan, has grown from $ 200 million at the end of 2019 to the current figure of $ 2.6 trillion.

Certainly, these digital assets are becoming a threat, but it will come from the voracious appetite for the minting and circulation of these currencies. Bitcoin and Ethereum, together, consume as much electrical energy per year than Indonesia. It leaves a generous carbon footprint as well as a growth electronic waste problem. Now that’s a concern.

RIDGEFIELD – A popular food truck that has run on Route 7 for more than a decade is looking for a new place to park after a site inspection found it was breaking regulations by the State.

The Zawack Shack normally parked on a freeway opposite the Days Inn by Wyndham, just a mile north of Branchville. It has sold lunch items to townspeople, truckers and travelers for 11 years and has become a staple restaurant during the pandemic as it is flanked by picnic tables.

But on October 7, state officials ordered Greg Zawacki to shut down amid the lunch rush. Zawacki’s mother, Joan, said they had had an hour to leave the site. The state Department of Transportation did not respond to requests for comment.

Grand Bassa County Electoral District No.5 representative Thomas Alexander Goshua vowed to do everything in his power to lobby his colleagues to ensure that funding for the establishment of the drug court goes through the national budget.

Representative Goshua revealed that his passion for lobbying for court funding is that drugs are becoming a serious “threat to national security and our future generations.”

The Grand Bassa County lawmaker noted that the tribunal is supposed to be a fast-track tribunal to provide a speedy trial for those accused of drug law violations if it meets the overwhelming support of the Senate Chamber.

“Those who are found guilty will go to jail for 20 years and more,” Goshua said. “I firmly believe that the drug court is the last alternative to deal with the problem of the drug influx into the country.”

“We can all agree that we cannot stop the proliferation of drug-related offenses that we experience and currently face in the country, in the absence of a specialized tribunal that will prosecute, arrest and incarcerate those on trial. guilty, ”Goshua said. “It will breathe new life into the country’s justice system and be an effective way to speed justice and eliminate delays in drug litigation.

According to Goshua, the special drug court will provide for a case to be dealt with and judged, and for a judgment to be rendered within a shorter period of time.

“This is one of the main ways to minimize drug offenses in the country, only if the government is interested in supporting the establishment of a tribunal,” Goshua said.

He also believes that stricter legal measures like the drug court should be put in place and with funding from the national government, stressing: “If we want to protect future generations and punish drug traffickers in our country, then we must all defend for the court. Goshua’s position on funding for the Drug Court comes days after members of the House of Representatives unanimously passed the Drugs and Substances Control Act of 2014.

It has been forwarded to the Senate and, if approved, will go to President George Weah for signature. decision of the House of Representatives at the time.

But, Representative Goshua said there were many loopholes in the Mulbah bill then, especially in the first against drugs and substances in the country.

“The current drug law of 2014 has a loophole in the fight against drugs and substances in our country. And it is the defect or the statutory infirmity that the amendment seeks to correct and give teeth to the law, ”noted Goshua.

During this time, he took the opportunity to call on all well-meaning citizens to join his advocacy and educate people about the danger associated with drug abuse. According to Representative Goshua, he will also lobby for funding to build a rehabilitation center for drug addicts, which would help reintegrate them into society to become better people. “I am going to press my colleagues to obtain budgetary support in order to make the competent institutions responsible for combating drugs in the country accountable. “

After its first nominee, a famous doctor, meets with fierce moral outrage, Los Angeles County Supervisor Kathryn Barger again tries to make a voice against the grain of the Los Angeles 10-member commission. Homeless Services Authority.

This time it might actually work.

On Tuesday, the Los Angeles County Supervisory Board will vote on Barger’s LAHSA candidate, Rev. Andy Bales, president and CEO of the Union Rescue Mission on Skid Row, which has for years denounced Los Angeles’ policy on homeless. Bales criticizes the dominant strategy known as ‘Housing First’, which aims to remove barriers such as sobriety requirements and move homeless people from the streets to single-family homes directly or after short stays in the city. shelter.

Barger said that while she supports the ‘housing first’ approach, she cannot address ‘everything that afflicts people on the streets’, and she believes Bales will bring a street-level perspective. to commission.

“The more people you have to provide solutions and thoughts, the better the system we will have to solve this problem,” said Barger, the only Republican on the supervisory board. “And I think having someone who disagrees isn’t a bad thing.”

Barger initially recommended Dr. David Drew Pinsky, more commonly known as “Dr. Drew”, to the LAHSA commission in April, but withdrew the nomination after massive opposition from homeless advocates.

Bales’s relationship with Barger dates back to 2007 when, as then assistant supervisor Mike Antonovich, she helped pave the way for the mission development of a women and children’s center in Sylmar. He calls him “a real hero in my eyes”.

Originally from Des Moines, Bales, 63, was teaching at a Christian school there in 1985 when he decided to devote his life to serving people living on the streets. He was ordained in the First Federated Church in Des Moines in 1989. Now living in Pasadena, he and his wife, Bonnie, a nurse, have raised six children and fostered 25 children in foster care. Bales, who has type 1 diabetes, lost his lower right leg to carnivorous infections he contracted on the streets of Skid Row and later had to have his lower left leg amputated.

He led other nonprofits in Des Moines and his current home, Pasadena, before taking over as director of the Union Rescue Mission in 2005.

Union Rescue Mission, founded in 1891 to distribute food and clothing from gospel wagons, is a private homeless service provider that today occupies a five-story building in the 500 block of South San Pedro Street , in the heart of Skid Row. He practices a faith-based “recovery model” providing immediate shelter, health care and life skills training for up to 1,000 clients.

According to his tax return, he offers an intensive 12-month program that includes 2,000 hours of 12-step Bible study, recovery classes, work therapy, counseling and fitness classes, followed by a 6 to 24 month transition program.

Under Bales’ leadership since 2005, Union Rescue Mission opened the Hope Gardens Family Center, the satellite in Sylmar for older women and mothers with children, and expanded its fundraising and strengthening mission of recovery in a Christian context. .

In 2011, Bales won his board of directors award endorsing the Gateway program to “instill a greater sense of responsibility and dignity” by charging a nominal fee for long-term shelter.

Bales criticizes the permanent housing strategy of leaving people on the streets for years because there is not enough housing and allowing what he sees as chaotic conditions when residents of those houses continue to live. consuming alcohol or drugs.

In an interview, he said he lovingly refers to the mission’s long-term housing “as our permanent supportive housing – the sober version – because we believe in creating environments where people can stay sober and recover in safe environments. And that’s one thing I want to point out, is that there is a missing link in the housing continuum that LAHSA currently has.

While this posture has made Bales an outsider in the secular nonprofit milieu of current homelessness policy, his undisputed passion and commitment to serving the homeless has isolated him from the wave of opposition that derailed Barger’s first pick from Pinsky.

In recent years Pinsky has argued that the homelessness crisis is not due to lack of housing but to mental illness and addiction.

Barger withdrew Pinsky’s nomination amid outcry – and rumors that his fellow supervisors would not support the nomination if passed.

Rarely are the appointments of supervisors to local councils controversial, and even rarer they do not receive unanimous approval.

The LAHSA Commission, which has the power to develop financial, planning and program policies, has 10 members, five appointed by county supervisors and the remaining five by the mayor of LA and city council.

It’s unclear how much power Bales might have as a single member, and even those who differ from him on strategy have refused to criticize his appointment, at least publicly.

Nan Roman, chief executive of the National Alliance to End Homelessness, did not criticize Bales’ appointment but suggested in an interview that it would not be helpful to have a commissioner arguing for a model special recovery.

She said it can be helpful for religious leaders ‘voices to be heard, but they need to be open to hearing data that shows their ideas, such as Bales’ support for transitional housing, might not not work for everyone. Research has shown that transitional housing is expensive and not as efficient as permanent housing.

Bales said he had no intention of disrupting.

“So the first step is to listen,” he said. “And second, you know, gently and humbly sharing my ideas so that they are welcome and at least heard.”

“I hope,” he added, “that there is room for a person of faith who believes in recovery and believes in immediate housing or shelter above everyone’s head. world in Los Angeles, not just a few, as is currently practiced in… Sometimes an opposing voice is needed to think things differently.

Bales backed Pinsky’s appointment and, when it was withdrawn, blamed the “culture cancellation” and “echo chamber” of Housing First and harm reduction advocates.

“It kind of told me that I wouldn’t be welcome either because I believe in recovery and in homeless people with addictions – whether the addiction developed because of it or led to it – the addiction still needs to be addressed in order for them to live a full, healthy life ”, Bales told Newsweek in April.

His appointment is expected to be approved at Tuesday’s supervisors meeting.

Addis Ababa, November 4, 2021 (ECA) – The United Nations Economic Commission for Africa (ECA) today hosted a high-level virtual consultative forum on the creation and establishment of the regional database on group purchases of selected drugs for maternal, newborn and child health. This consultative forum follows the successful launch of the Centralized Procurement Mechanism (CPPM) of the AfCFTA-anchored Pharmaceuticals Initiative which was held on July 29, 2021.

The objective of the forum was to advocate for the creation and implementation of the electronic group purchasing database platform, to review the list of maternal, newborn and child health products, as well as to raise awareness on the roles of pilot countries regarding the operationalization of the CPPM. The meeting provided a platform to discuss the creation of the Pooled Procurement Network (PPN), a platform that enables staff involved in the procurement of health products to share information about the procurement process.

During the opening speech, Ms Jane Karonga, head of the AfCFTA-anchored Pharmaceuticals Initiative project, said the global response to the Covid-19 pandemic has revealed gaps and gaps in the supply chain and health care systems. “Even though the AfCFTA-anchored Pharmaceutical Initiative was launched a year before the pandemic hit the world, the groundbreaking work that has been undertaken on it has shown the potential of what countries can achieve if they Leverage AfCFTA and WADA to scale up local pharmaceutical activities, influence prices with pooled supply agreements and ensure harmonized regulatory framework and quality standards, she said. concluded by noting that the three pillars of the project were put to good use in the creation of spin-off institutions such as the African Vaccine Acquisition Trust (AVAT) and the African Medical Supplies Platform (AMSP), the first pooled procurement mechanism of the platform. online form owned by Africans in Africa, are real testimonies to the success of the initiative.

The advisory forum brought together senior government officials, procurement experts and ministry of health focal points from the ten pilot countries and a team of continental experts.

Pilot Member States unanimously agreed that the concept of the Regional Maternal, Newborn and Child Health Medicines Database will be a game-changer for the selected pilot countries and by nature the continent. The operationalization of the database will create an enabling environment for the pooling of procurement processes in order to attract favorable prices in the short to medium term, which in turn will create the fiscal space essential for countries to develop. more the health sector or any other sector of the economy. They also agreed that alignment of national stakeholders is essential for successful implementation.

At the end of the forum, Member States appreciated the consultation and looked forward to future commitments on the implementation of the Pharmaceuticals Initiative anchored in AfCFTA.

ISLAMABAD: Foreign Minister Shah Mahmood Qureshi on Tuesday expressed hope that the signing of a Pakistan-Uzbekistan protocol on the establishment of a Joint Security Commission will help further strengthen relations between the two countries.

Qureshi was addressing the Secretary of the Security Council of Uzbekistan, Lieutenant General Victor Makhmudov, who held a meeting with him here at the Foreign Ministry.

During the meeting, they discussed issues of mutual interest, including bilateral relations, defense and military cooperation, and developments in Afghanistan, according to a foreign ministry statement.

The two sides also reaffirmed to activate the existing defense and military cooperation agreements between Pakistan and Uzbekistan.

Foreign Minister Qureshi said Pakistan attaches special importance to its bilateral relations with Uzbekistan, adding that relations between the two countries are based on brotherly ties, common religious and cultural values.

He said Pakistan was seeking to transform long-standing relations with Uzbekistan into a strategic partnership.

He said Prime Minister Imran Khan’s visit to Tashkent on July 16-17, 2021 and his meeting with the Uzbek president further strengthened bilateral ties.

Pakistan, as part of the “Vision Central Asia” policy, seeks to promote ties with Central Asian states, including Uzbekistan, he added.

He hopes that the agreement signed Tuesday between Pakistan and Uzbekistan on the establishment of a joint security commission will further strengthen relations between the two countries.

He said the commission will help prevent terrorism, drug trafficking and also provide a ground for the two countries to cooperate with each other during natural disasters.

Regarding Afghanistan, the foreign minister said Pakistan and Uzbekistan have similar views on the situation in this war-torn country.

Qureshi called on the international community to help the Afghan people to resolve the humanitarian crisis in Afghanistan.

The foreign minister further said that Pakistan is focusing on its geopolitical priorities, adding that peace in Afghanistan will strengthen trade and economic ties with Central Asian states.

As Afghanistan’s closest neighbors, Pakistan and Uzbekistan have a common interest in bringing peace to Afghanistan.
